Leibovitz’s L-15 Medium was initially developed for use in CO2-free environments and therefore does not include sodium bicarbonate in its formulation. Instead, the medium’s pH is buffered through the use of salts, freebase amino acids, and galactose as a substitute for glucose. This helps to maintain the physiological pH of the solution. Leibovitz’s L-15 Medium is capable of supporting various cell lines, such as HEp-2 and LLC-MK2, as well as primary embryonic and human tissue cultures.

Reconstitution: Usually the medium is supplemented with 0.3 g/l L-glutamine (2 mM).
